Plasmid and lambda DNA are pre-digested with restriction enzymes - endonucleases that recognize and cut double-stranded DNA within or near defined base sequences. Digests are separated by agarose gel electrophoresis.

• Introduce restriction enzymes and their ability to digest DNA at specific nucleotide sequences
• Understand how electrophoresis separates differently sized molecules
• Perform agarose gel electrophoresis
• Load, run, analyze, and compare six restriction enzyme cleavage products (uncut, EcoRI digested, BglI digested lambda DNA and plasmid DNA)
