A specific mRNA is reverse transcribed to double-stranded DNA. This DNA product is then amplified by PCR. This reaction demonstrates the mode of replication of HIV, which contains reverse transcriptase. This experiment is the first introduction of a commercial RNA experiment for the classroom laboratory.

Group size: For 6 lab groups

Time required:

PCR Setup - 10 minutes
PCR - 3 hours
Electrophoresis - 60 minutes
Staining - 5 minutes

Kit includes: Instructions, RNA Template Concentrate, Primer Mix Concentrate (2 primers), Tubes with RT-PCR EdvoBeads™, RNAse-free water, EdvoQuick™ DNA Ladder, UltraSpec-Agarose™, 50X Electrophoresis Buffer, 10X Gel Loading Solution, InstaStain® Ethidium Bromide, Microcentrifuge & PCR Tubes, & Wax Beads.

All you need: DNA Electrophoresis, Thermal Cycler, Micropipets: 5-50 µl, UV or Blue Light Transilluminator, (3) Water Baths (if performing experiment without a thermal cycler), Microcentrifuge & Microwave or Hot plate.
